We explored the potential of inositol hexakisphosphate (IP6) and its analogues as a treatment for calcium oxalate (CaOx) nephropathies, conditions often linked to kidney stones. Our study utilized a new machine learning approach that helped screen various chemical modifications of IP6 to find a particularly effective compound.
This innovative method led us to identify a divalent inositol phosphate that demonstrated remarkable activity. In our findings, this molecule completely inhibited the crystallization of CaOx crystals at very low concentrations. Additionally, it significantly reduced the interaction between crystals and kidney cells, preventing harmful changes in gene expression related to kidney damage.
We observed that, in a mouse model experiencing increased levels of oxalate, the IP6 analogue reduced crystal formation and subsequent kidney injury. With these promising results, we believe that these inositol analogues may offer a new therapeutic option for those suffering from calcium oxalate-related kidney issues.
